Here are some possibilities:

If the curve family does not represent a range of average gradient values that includes 0.62, or if you have neglected to enter an ISO value in the film data entry window, or if you have chosen to include flare in the curve family (without first having run through the analysis far enough to establish the speed point location), the ISO buttons will be dimmed and disabled because the ISO reference curve can’t be identified under these conditions.
