It's difficult to describe metering techniques (much easier to demonstrate them) but in general the high reading is taken in full light and the low reading is taken in open shade. Just where in the shade depends on how much "fill" light is present, the reflectance of the subject, and how you want to interpret the subject.
Closeups of small objects are more difficult to meter but the incident meter isn't interested in the subject itself, so you can establish a satisfactory light arrangement, then remove the subject temporarily while you take the meter readings.
